Strain Description
                            
                                TIGIT (T cell immunoreceptor with Ig and ITIM domains) is a key immune checkpoint protein that plays a critical role in immune regulation and tumor immunology. By binding to its ligand CD155, TIGIT effectively suppresses the activation of immune cells such as NK cells and T cells. Furthermore, TIGIT exhibits synergistic inhibitory effects with other immune checkpoints, such as PD-1/PD-L1. Dual blockade of TIGIT and PD-1 can restore the co-stimulatory function of CD226, thereby enhancing the anti-tumor activity of CD8⁺ T cells. This mechanism underscores the significant potential of TIGIT inhibitors in treating various advanced cancers, including solid tumors such as lung cancer, gastric cancer, melanoma, and lymphoma, as well as hematological malignancies like multiple myeloma.
MingCeler Biotech has independently developed a TIGIT humanized mouse model by replacing the extracellular domain sequence of the TIGIT gene in C57BL/6N mice with the corresponding human sequence. This model retains the intracellular portion of the murine TIGIT protein, ensuring normal intracellular signal transduction and maintaining physiological functions. The TIGIT-humanized mouse serves as a valuable tool and an excellent platform for studying immune- and tumor-related diseases centered on the TIGIT protein.
